چکیده

Chlorotetracycline (CTC) has been used in many cells as a probe for membranous calcium. In polymorphonuclear neutrophils (PMN). the changes in CTC fluorescence upon stimulation are considered to monitor an early event in the activation process. Using quantitative video-enhanced microscopy. we report that in resting cells about 80% of the CTC signal emanates from the perinuclear region of the cell. indicating that internal structures are labeled with CTC. Approximately 20% of the total CTC fluorescence is taken up in a compartment sensitive to mitochondrial

Intracellular calcium storage and release in the human platelet. Chlorotetracycline as a continuous monitor.

The calcium-sensitive fluorescent probe chlorotetracycline was used to monitor calcium movement in human platelets. The chlorotetracycline fluorescence signal is a linear measure of the level of free calcium in the dense tubules and in the mitochondria, with probe sensitivity in the millimolar range. Experiments perturbing the system with the calcium ionophore A23187 shows that the level of fre...

Evidence for an association between Wnt-independent -catenin intracellular localization and ovarian apoptotic events in normal and PCO-induced rat ovary

The association of secreted frizzled related protein type 4 (Sfrp4) as an antagonist of Wnt mole-cules in apoptotic events has been reported previously. Moreover, its increased expression has been reported in the ovary of women with polycystic ovary (PCO). We have demonstrated in-creased Sfrp4 in PCO-induced rat ovary related to an increased number of apoptotic follicles showing nuclear ?cateni...

Involvement of membrane calcium in the response of rabbit neutrophils to chemotactic factors as evidenced by the fluorescence of chlorotetracycline

We have utilized the fluorescent chelate probe chlorotetracycline to investigate the possible involvement of membrane calcium in the response of rabbit peritoneal neutrophils to chemotactic factors. Two chemotactic factors, the small molecular weight fragment of the fifth component of complement C5a and the synthetic peptide formyl-methionyl-leucyl-phenylalanine (F-Met-Leu-Phe), were tested and...

Intracellular divalent cation release in pancreatic acinar cells during stimulus-secretion coupling. II. Subcellular localization of the fluorescent probe chlorotetracycline

Subcellular distribution of the divalent cation-sensitive probe chlorotetracycline (CTC) was observed by fluorescence microscopy in isolated pancreatic acinar cells, dissociated hepatocytes, rod photoreceptors, and erythrocytes. In each cell type, areas containing membranes fluoresced intensely while areas containing no membranes (nuclei and zymogen granules) were not fluorescent. Cell compartm...

Comparative Studies of High Contrast Fluorescence Imaging Efficiency of Silica-coated CdSe Quantum Dots with Green and Red Emission

Herein we report the possibility of using green and red emitting silica-coated cadmium selenide (CdSe) quantum dots (QDs) for remarkable stem and cancer cellular imaging, efficient cellular uptake and fluorescence imaging of semi and ultra-thin sections of tumor for in vivo tumor targeted imaging applications. The comparative studies of high contrast cellular imaging behaviours of the silica-co...
